As the COVID-19 pandemic spread across the USA in early 2020, many facilities had to reduce operations, preventing auditors from traveling to customer sites. Alturas Analytics developed processes to permit auditors to conduct their inspections remotely. Additionally, Alturas Analytics was one of the first companies to be inspected remotely by the FDA in April. This presentation will present tips for a simple and positive remote audit experience and describe the process employed by the FDA to successfully complete a remote regulatory inspection.

Microsampling of Non-Invasive Matrices: Practical Examples Using Tears and a Perspective of Past and Emerging Technologies

Presented by Jason Watts, PhD, virtually at the AAPS Land O' Lakes Bioanalytical Conference. 

Non-invasive matrices such as tears, sweat, saliva, and milk offer an enticing alternative to traditional sampling from blood, serum, and plasma.  Collection of these matrices is simpler and more affordable than venipuncture, as well as being much less unpleasant for patients and study subjects allowing for more frequent sampling, higher compliance, and removal of fear of needles as a barrier to clinical trial recruitment. However, bioanalysis of these matrices is not without challenges. Sample volumes can be very low, the more complex matrices such as milk may require extensive sample preparation, and the use of the matrix must be biologically relevant. Using the example of a method we developed at Alturas Analytics Inc. to measure Tobramycin from human tears, we will discuss practical applications of microsampling of non-invasive matrices for bioanalysis, and improvements offered by emerging technologies.

Scientist Focus: Jason Watts, Ph.D.

Jason Watts, Ph.D., is a senior scientist at Alturas Analytics, Inc., focusing on HPLC-MS/MS techniques for method development, validation, and sample analysis of new chemical entities and biomolecules.  A diverse background in molecular genetics and animal nutrition, coupled with over ten years of applied mass spectrometry experience, provides the expertise vital to delivery of innovative solutions to bioanalytical challenges.

What inspired you to become a scientist?
My worldview has been greatly shaped by my love of Star Trek: The Next Generation. This is a world in which problems are solved by reason  and the scientific method. Becoming a scientist was the logical thing to do.

Did anything make science memorable for you when you were younger?  
When I was a child everyone I knew got chickenpox.  When a kid you knew got it, our families would throw a party to try to expose their children. It was a madness that we all just accepted as a normal part of life. And then the vaccine became widespread and it just stopped. Many people my age and older are covered in scars. A few years younger than me, nothing. Science did that. 

What inspires you?
Everyday I get to work with a bunch of people who are a lot smarter than me. It's great!

What are your favorite studies to analyze?
I like a challenge. At Alturas, we often get projects that other companies couldn't get to work. Getting a method with a low limit of quantitation to work for a tough analyte...very satisfying.

Why is science cool?
Learning and curiosity are the basis of science. Alexander Fleming's curiosity about the mold growing on his bacterial plates led to penicillin saving untold millions of lives. I think that is pretty cool.

What is the most rewarding part of your job?
We get to play a small, but critical role in developing truly novel treatments. We work with truly innovative clients and our combined efforts have tangible benefits for people.

Alturas QAU Provides Virtual Audits and Consistency During COVID-19

April 17, 2020

As research continues through the COVID-19 pandemic, the need to continue operations as usual and manage critical timelines within regulatory compliance presents unprecedented challenges for the entire pharmaceutical industry. Due to the potential spread of COVID-19, the Alturas Quality Assurance Unit (QAU), working with Alturas’ IT, introduced the ability to conduct virtual audits. These audits include live web-based discussions, in-depth interactive facility tours and secure folder access for file sharing of controlled documents in strict adherence with Alturas SOPs.

Alturas’ QAU is taking additional in-house precautions to further conform to CDC recommendations. In-process critical phase audits of regulated studies are being conducted as scheduled while working within the recommended social distancing guidelines. To reduce opportunities of possible COVID-19 spread, the time spent with Alturas scientists during inspections and within the QAU group is minimized through working remotely and staggering work schedules.

Alturas’ QAU continues to address and implement comprehensive SOPs to provide structure and consistency for all operations of Alturas Analytics during the changing landscape of COVID-19.  While Alturas remains open as an essential business, the QAU ensures that pre-clinical and clinical study activities are conducted in compliance with procedures and applicable regulations on schedule.

An Application of Scientific Research in the Private Sector: Supporting Novel Drug Development on the Palouse

Alturas IT Make Critical Changes to Support Business Continuity During Pandemic

April 2, 2020

Alturas’ Information Technology department has made critical and expedient changes to support our business continuity plan and adapt to the changing workflow influenced by the coronavirus. To enable more employees to work from home, we have doubled our internet bandwidth and acquired additional VPN authentication devices to accommodate all off-site employees. New collaboration apps are being integrated to maintain communication, efficiency, and workflow continuity for dispersed employees. We have also purchased new computers and updated lesser used workstations for employee use at home.

The IT staff have moved to a coordinated schedule in order to offset shifts and minimize personal contact while still providing the necessary support for employees working irregular hours or weekends. Support practices have also been modified to further reduce personal contact and cross-contamination of computer equipment. 

Alturas has accelerated the deployment of IT infrastructure at our future branch office. If needed, this space will provide additional workspaces to further reduce contact between personnel.

As Alturas’ IT Department ramps up capacity for working remotely, and provides more tools for communication and collaboration, relevant security and compliance policies are continuously updated to ensure the data safety and security of client data.  All work being completed remotely remains in accordance with Alturas’ Standard Operating Procedures. 

With ongoing changes during this pandemic, Alturas is committed to providing clients, sponsors, and collaborators with the same secure on-time, expert bioanalytical services. Our full operation is maintained with no negative impact to services or schedule.

Alturas continues to closely monitor developments related to the COVID-19 outbreak and its impact to our location, services, and supply chain.  

  • Alturas Analytics is considered an essential business and will remain open during “Stay At Home” orders imposed through city, state, and federal regulations. 

  • Due to a decision to limit visitors at our facility, virtual audits are now available and can be scheduled through QA Manager David Schumacher, RQAP-GLP.

  • Resources are focused on Sponsor projects to stay ahead of schedule while providing the option for personnel to work remotely in order to maintain workflow continuity. 

  • Spare parts, excess supplies, and consumables are retained on site to ensure no supply chain interruptions.

  • With ongoing closures of global laboratories, Alturas is still in full operation and capable of supporting all bioanalytical services offered in our standard portfolio.

Alturas Analytics Uninterrupted During COVID-19 with Integrated Data Management Software

March 25, 2020

In 2018, Alturas launched a fully integrated real-time data management system to streamline all systems necessary for bioanalytical laboratory operations with high visibility, productivity and efficiency. This database, the Alturas Laboratory Interface Communications Equipment, or ALICE, has proven to be an invaluable tool during the current coronavirus pandemic.

The impact of isolation during this time can pose difficulties if communication, lab supplies, equipment lists, test methods, project management, and invoicing are managed in separate locations with individualized design and use. Using ALICE, Alturas has not faced these traditional difficulties and has maintained the ability to continue focusing on sponsor projects. Alturas has been able to stay ahead of schedule to better serve our clients while providing the option for personnel critical to the operation to work remotely, maintaining workflow continuity.

The standardized and customized software enhances remote work through quick and easy access to current project metrics including status, laboratory activities, batch runs, invoices, contracts with real-time visibility. Forecasting project timelines and potential bottlenecks has even greater importance during these unforeseen circumstances. Prudent resource management based on real-time data ultimately provides security and assurances throughout all laboratory operations.

While Alturas is following CDC guidelines to protect the health of employees, ALICE is reducing disruption to Alturas’ business continuity. This custom built, integrated project management suite is transforming how our business can be conducted remotely indicating safety and health measures of COVID-19 will not provoke any insurmountable obstacles regarding business operations.

Further cross-training between functions and across projects has been implemented in anticipation of a possible reduction in workforce as the situation evolves. With the infection rate low in our region, Alturas intends to operate cautiously under strict safety and health measures that will minimize the risk and spread of illness among staff.

Current Status and Ongoing Response to COVID-19 - Updated 18Mar2020

Alturas Analytics, Inc. is actively participating in social distancing by encouraging those who can work from home to do so.  Alturas has also mandated that any employee who has traveled recently or will be traveling to an outbreak location must self-isolate for 14 days before returning to the facilities.  These employees are encouraged to continue working from home. 

Alturas is able to carry out this strategy as a result of its preparedness plan.  Extra bandwidth and equipment, supplies, spare parts, and consumables are stored on-site to minimize interruptions.   A fully integrated real-time data management system connects all operational workflows and processes. This allows full visibility of project status including timelines, review processes, reporting, batch runs, invoices, contracts and real-time alerts under secure remote access.  

A system for conducting virtual audits is in final test to ease disturbances and avoid delays caused by social distance and travel restrictions. 

Alturas’ operation remains consistent and will post updates regularly as circumstances change.

At Alturas Analytics, Inc. the health and safety of our sponsors, employees, stakeholders, and all those Alturas serves is our top priority.  Alturas is closely monitoring developments related to the COVID-19 outbreak and its impact to our location, services, and supply chain.  

Alturas is taking appropriate and necessary preventative measures to minimize the risk and spread of the illness. Following Centers for Disease Control and Prevention (CDC) and regional health organization recommendations, a number of precautions to preserve health and safety have been adopted.  

EMPLOYEES

Alturas encourages healthy behaviors of its employees.  Precautions such as additional hand washing stations throughout the facility and elimination of business and social gatherings such as conferences, seminars and workshops, as well as gathering within the facility, are in effect.  Universal health precautions are a requirement in the laboratory environment and strictly enforced at Alturas. 

As part of Alturas’ routine operations, within Alturas’ business continuity plan, cross training for essential positions has taken place in case of a reduction in workforce.  Alturas has also taken steps to reduce viral load exposure to help with recovery process, and prepared with additional equipment and bandwidth for at risk employees to work from home.  It is mandatory for employees to stay home if feeling ill.

FACILITIES

Alturas is committed to providing our clients, sponsors, and collaborators with the same on-time, expert bioanalytical services. We do not foresee any interruptions in scheduled maintenance for equipment. In addition, a supply chain awareness and preparedness plan was implemented in January to ensure the supply chain remains uninterrupted.  Furthermore, extra spare parts, excess supplies, and consumables have been retained on site.

At this time, Alturas is open for audits, but this could change as the situation evolves.  For now, if you do choose to visit Alturas’ location, please know sanitation efforts are increased and steps have been taken to reduce exposure to concentrated viral loads by offsetting shifts and work days and lowering building temperature.

The 2018 FDA Guidance for the Validation of Bioanalytical Methods requires an assessment of the impact of the presence of co-administered drugs on the analytical measurement and stability of novel drug candidates. GLP-compliant methods were developed and validated in support of a clinical study that featured co-administration of itraconazole, paroxetine, cimetidine, moxifloxacin, and valproic acid with a proprietary drug candidate. Various methods involving protein-precipitation and liquid-liquid extraction were employed utilizing direct-analysis or dilution of supernatant as well as evaporation with nitrogen followed by reconstitution of analyte in organic solvent. Read the Abstract

Contract research organizations (CROs), pharmaceutical companies and regulators have extensively discussed co-administered stability since it was brought up in 2010 at the Fourth Calibration and Validation Group Workshop on Recent Issues in Regulated Bioanalysis. The questions are whether or not co-administered drugs impact the stability of an analyte, and what regulatory requirements should exist for cases of co-administered drugs. Read More

Jennifer Zimmer, Ph.D. has been with Alturas Analytics since 2005. She has a vast background in bioanalytical as well as pharmacological science. As Laboratory Director, Dr. Zimmer is responsible for the overall operation and technical oversight of producing quality assays, generation of data and reports of the highest integrity and adapting to the changing landscape of the industry. She is an active participant in the Global CRO Council but is most proud of mentoring a team of scientists to be successful contributors in the bioanalytical industry.   

Dr. Zimmer received her B.A. degree in English and Zoology from the University of Idaho and her Ph.D. in Pharmacology from the University of Colorado Health Sciences Center.

What is the most rewarding part of your job?

Getting to see the impact that the novel compounds that we help analyze can have on the quality of life for patients. Even though our piece of the drug development process is a small one, it is really neat to see that the work we do at Alturas has a real benefit.

What accomplishments are you most proud of?

I am proud of how we have developed our training program to help us really build our team here at Alturas. The senior members of the team have worked together to make our training program build from the bottom up, which allows for a more seamless process and to helps all of our newer employees to be as successful as possible.

 What is the most challenging aspect of your job?

The ever-shifting timelines of drug development. This is also sometimes a fun challenge to see how we can best arrange timelines to make sure that we meet all of our sponsor’s deadlines.

Effective July 16, 2019, Alturas Analytics is thrilled to announce the appointment of Dr. Lori Payne to the Business Development team as Executive Director of Business Development joining Ann Hoffman, VP Strategic Development.

 

"Dr. Payne brings a wealth of business development and laboratory operational experience to Alturas. She is well known in the field of bioanalytical and pharmacokinetic analysis for both large and small molecules as well as immunogenicity," says Ann Hoffman. Dr. Payne's focus will be developing new business opportunities in the bioanalysis of large and small molecules using LC-MS and innovative technologies.

 

Dr. Payne has had a distinguished career in large pharma as well as managing and directing growth and development in Contract Research Organizations. She advanced steadily in her career at Merck from Senior Research Chemist to Group Leader and Senior Scientist. As Senior Scientist, she led a group of 8 PhD/MS/BS level PKDM department scientists in all aspects of project management, drug development, and bioanalytical analysis of pharmaceutical drugs using LC-MS and other analytical techniques. She subsequently joined the team at BASi (formerly LC Resources) to lead their West Coast Operations as Laboratory Director and General Manager. The site achieved numerous awards for their lean and continuous improvement focus. Dr. Payne was named “Manufacturing Leader of the Year” by the McMinnville  Economic Development Partnership. When the site was consolidated with two others, she was selected as Vice President to run the Analytical, Bioanalytical and Discovery Departments; a team of about 65 staff including quality assurance, sample management, and business development functions. Dr. Payne furthered her Business Development skills and strategies as the Director of the US Pharma BD Teams at Intertek.

Dr. Payne earned a PhD in Chemistry from LSU Baton Rouge and a BS in Biochemistry at UC Davis.

 

She says, “I’m honored to join the Alturas team which has an excellent reputation for quality work and superior science. I’m excited to merge my business skills with such a well-run continuous improvement minded laboratory with a heart.”

Abstract: Antibody drug conjugates (ADCs) are potent and specific biopharmaceuticals. These large molecules are typically analyzed using ELISA methods, which do not have the selectivity to accurately quantify the ADC alone. HPLC-MS/MS analysis of the digested ADC is an alternative approach that provides better selectivity. In order to increase the selectivity/sensitivity of the method, analysis was conducted using microflow HPLC (MFLC) and immunocapture sample preparation. Read Full Abstract

Chad Christianson, Senior Scientist

Chad has 20 years of related experience as an analytical scientist with over 15 focused on bioanalysis at Alturas Analytics. This depth of experience in applied LC-MS/MS and GC-MS/MS, along with an education in chemical engineering provides the backbone for productive and innovative science.  Chad leads the protein quantitation group at Alturas, applying novel techniques to a regulated, high-throughput production environment.

What inspired you to become a scientist?
Learning about the scientific method in grade school intrigued me. Observe, make a hypothesis, test the hypothesis and draw a conclusion/change hypothesis. I’ve always wanted to know why something worked and this method laid the groundwork for my career.

Did anything make science memorable for you when you were younger?
Most definitely. My exposure to science fiction books, television and movies had a huge impact on my realization of the possibilities of science.

What is the most challenging aspect of your job?
Developing novel methods for compounds that have stability, sensitivity and selectivity challenges.  

What inspires you in the bioanalytical industry?
Playing a small part in the approval of drugs and therapeutics biologicals that can transform the lives of people suffering from disorders previously untreatable.

What are you favorite studies to analyze?
Phase 3 Clinical studies because based on these results the possibility of approval is in sight or it is time to start working on the method for the backup compound.   

What accomplishments are you most proud of?
Watching Alturas blossom into a real force in the bioanalytical community. When I started in 2004 we only had three full time employees and two instruments. Now we have 40 employees, 18 instruments and have the reputation of taking on projects that have failed elsewhere or are too difficult for most labs to handle. We have a fantastic team assembled and I am honored to be part of it.

Why is science cool?
Science allows an individual to ask a question, research/experiment and come up with an answer. Sometimes that answer isn’t what is expected but the knowledge obtained getting to the answer can be a tool used for the rest of your life.

What is the most rewarding part of your job?
Seeing people (especially children) with little hope for treatment receive a second chance. We have the honor and privilege of making a positive impact on the world.    

Do you have any advice for people considering careers in a bioanalytical laboratory?
Be prepared to embrace failure as a learning tool. The knowledge of knowing what doesn’t work makes it much easier to discover what does work and why.

“Pharmacokinetic studies are a critical component in evaluating the safety and efficacy of a drug from discovery through post-marketing clinical trials.” Sara Underwood spent 16 years in the biotechnology industry performing pharmacokinetic analysis in support of preclinical and clinical drug development of small molecules and biologics. Sara joined the Alturas team in 2014 bringing PK/TK expertise in-house as a strong complement to the bioanalytical services provided at Alturas.

Sara brings the knowledge as a scientist and a unique perspective from the biotech industry that has greatly contributed to the growth and success of Alturas Analytics. When not at work, Sara may be found galloping her horse over the Palouse, or hiking with her husband and son.

What inspired you to become a scientist?

Growing up I was always asking the question ‘Why’? My sophomore year in high school was when I decided I wanted to study biology so I could finally get answers to all of my questions.

What accomplishments are you most proud of?

Balancing multiple projects while not sacrificing quality.

What is the best – and the most challenging – about PK/TK analysis?

The best part is having a role in moving a project forward through the development process. The challenging part is only having a glimpse of that project.

What is the most rewarding part of your job?

Working with a coherent team who genuinely care about the quality of work and helping to support one another in meeting the project goals together.

A Message from Robin Woods, Alturas Analytics President

 

As the sun sets on a successful 2018, and we open 2019 feeling invigorated and are poised for another growth year at Alturas, we would like to thank our partners for your continued confidence in and commitment to the Alturas team.

 

Most exciting to share is that Alturas is entering the planning stages for an expansion that will more than double our size. We have identified and placed a purchase option on land adjacent to our current building. We have also hired an architect, assembled a building planning committee and are refining layout ideas to build a state-of-the-art facility that is specifically designed to be a streamlined, efficient bioanalytical lab.

 

As a continuation of our company’s development, we have increased our offerings in support of regulated studies. Microflow and hybrid LC-MS/MS techniques are now fully engaged in routine analysis that include new biological entities, biomarkers, ADCs, as well as small peptides and new chemical entities. We are also very pleased to utilize in-house expertise in pharmacokinetic (PK) and toxicokinetic (TK) analysis by offering PK and TK reporting in support of studies from discover through clinical trials.

 


 

Alturas is also proud to support local non-profits and we are fortunate enough to be able to give back to our community that has supported us for more than 18 years! Here are a few of the organizations we supported this year:

 

  • Science on Ice  – A great field trip opportunity for schools and community groups that combines science lessons, physical education, and ice skating fun.

  • The Gritman Foundation - The Gritman Foundation is the philanthropic partner of Gritman Medical Center, helping Gritman fulfill its mission of providing extraordinary care to the people in our region.

  • Christmas for Kids - A local non-profit organization providing Christmas gifts and warm clothing for the children of low-income families in Latah County.

  • Alternatives to Violence of the Palouse - An organization that supports victims and survivors of violence in the Palouse region.

A Message From Robin Woods, President

At Alturas Analytics, 2016 was the year for building infrastructure and preparing for growth to meet new challenges. With investments in capital equipment, scientific staff, a facility expansion, and company-wide software upgrade and integration, we enter 2017 with greater capacity and streamlined processes operated by an enhanced team of talented professionals.

The Year Ahead

With gains in sensitivity from a wide variety of analytical platforms in combination with the specificity of immunoaffinity capture, we anticipate an increase in the migration of large molecule quantitation to LC-MS/MS from traditional techniques. Alturas is engaged in a focused effort in this area to provide fully validated assays for biologics, whether protein and peptide therapeutics, biomarkers, antibody-drug conjugates (ADC) or RNA drugs to address the non-specificity and often lengthy development cycles of ligand binding assays. Routine LC-MS/MS bioanalysis of large molecules performed in a regulated environment is a great complement to our lengthy history and extensive experience with ADCs. With our scientific expertise and the loyalty of our ADC clients, this presents major growth potential for Alturas.

As a pioneer of dried matrix spot analytical procedures, we are encouraged with the resurgence of innovative developments in microsampling techniques as a viable alternative to standard blood draws. Collection cards, capillaries, swabs or other microsampling techniques are ideal when sample volume is limited such as with preclinical models, tears, support of pediatric studies or collecting samples in remote regions where centrifuges and freezers are scarce or nonexistent. Alturas remains an active contributor in the realm of microsampling techniques and we are currently working towards the use of NoviplexTM plasma extraction cards in support of regulated bioanalysis.  Unlike dried blood spot cards, plasma is extracted from a single blood drop so hematocrit effects do not exist, samples are more stable, and data is comparable with methods that use conventional plasma collection procedures. Microsampling is already embedded into routine preclinical workflows in the research environment. 

Alturas has much to look forward to in 2017 and beyond with enhanced, streamlined operations and a deepened portfolio. Our timeline driven service strategy provides the culture for controlled growth and stability to succeed in a progressive industry that is constantly pressing the limits
